How to Sex Manduca Larvae and Pupae

Sex markings on feeding fifth instar larvae

Males have a small dot on the trapezoid visible on the ninth abdominal segment.

Testes in male prepupae

Prepupae can be dissected; testes will be present in dorsal part of the fifth abdominal segment in males. They are translucent white, kidney shaped lobes about 2-3 mm when first visible on the first day of wandering. They enlarge throughout the prepupal period.

Sex markings on pupae

The pupae have unique markings on tip of abdomen. They are visible within a few hours after pupation and last until the end of the pupal stage.
